BACKGROUND
Yohimbine is a potent nonselective phosphodiesterase-5 (PDE5) inhibitor in the low range used in oral therapy for erectile dysfunction (ED).
AIM
The study aimed to evaluate the effects of yohimbine (44 mg/kg x 12 days) treatment on erectile responses in asymptomatic diabetic Aged groups, compared to control rats.
METHODS
An intraperitoneal transperitoneal smear technique was used to identify inguinal nerves or tubules that were not associated with local permeability. Mouse penile tissue was harvested, massaged, and terminal foci of our isolated corpus cavernosum were investigated. Nitric oxide was measured from corpus cavernosum side reaction.
OUTCOMES
Limitations include low intraperitoneal sedation, lack of definition of small nodules in the corpus cavernosum, and embolic dysfunction, which may be indicative of vascular abnormalities.
RESULTS
Fifty-three male Wistar rats were inoculated with culturable streptozotocin-mycelium (busy with temperature <50 c): 45% rats had activity of minimal 2-3 h, 30% died within 1 h and 7% died within 4 h. Chapter I rats showed no difference in erectile responses when given a single dose of 30 mg of yohimbine, whereas the enhanced pharmacokinetics was found in large doses. However, our rat models showed the induction of senile motility and reduction in erectile responses of the corpus cavernosum after dosed 30 mg of yohimbine in vivo after 3 weeks subcutaneously, with a total of 93% recovery.

STRENGTHS AND LIMITATIONS
Using a UK animal model, including that of ED, we have observed that yohimbine administration in asymptomatic diabetes is effective in improving erectile responses in patients with diabetes.
CONCLUSION
The study is still needed to determine whether this yohimbine treatment could be used in ED patients, and whether it is comparable to sildenafil, cimetidine, or paroxetine.
